
The HistoSonda protocol is simple. It requires neither special apparatus nor working solutions different than those of standard immunohistochemistry.
This protocol has been optimized for formalin fixed paraffin embedded tissue sections of 4-6µm thickness. Tissue sections must be adhered to silanized microscope slides.
The DNA probes are dissolved in a very viscose hybridization buffer that contains elevated concentrations of high molecular weight dextran. For this reason the first step in the protocol should be to reconstitute the probe.

* Probe Incubation: 60 min
* Primary Antibody: 30 min
* Secondary Polymer Antibody : 20-30 min
* DAB Revealing: 5 min
HistoSonda® hybridizes in just 60 minutes at 62 ºC - the fastest available incubation for in situ hybridization probes.
The whole protocol from dewaxing to revealing can be performed in only 3 hours, most other probes require overnight incubation and are long and complicated.
